I suspect other strong contenders will be Andrew Burnap and Paul Hilton (‘The Inheritance’), Mark Addy and Gabby French (‘Hangmen’), Andrea Macaset, Brittney Mack, and Samantha Pauly (‘SIX’), Ben McKenzie (‘Grand Horizons’), Chantal Thuy (‘Linda Vista’), and Dharon E. Jones (‘West Side Story’).
I believe they have slightly bent the rules before to allow someone to receive this award if their only other credit was an understudy or replacement (since they didn’t debut the character), as the rules now read “for their significant debut performance”. If they go that route then Lauren Patten (‘Jagged Little Pill’) could be a good choice (her one previous bway credit is an understudy/replacement for ‘Fun Home’).

I saw them in person the past three years, and in 2018 I even went to the afterparty. It was a lot of fun joining a bunch of fellow theatre nerds singing “Out Tonight” and “My Strongest Suit.”
